Brain-like multi-layer tissue models are in great needs for the study of neurological diseases for which they will be less different from human brain compared with animal models and have a higher impact on the study of diseases. However, there are many limitations for the conventional tissue engineering approaches due to lack of flexibility in positioning cells, falling short in the ability to pattern cells into complex 3D architectures and lack of the cell culture environments control; The emerging 3D cell printing technology can handle these perfectly. Whereupon, a printing and culturing system, integrating cell printing platform and cell culturing system, was designed and fabricated to achieve the biofabrication of multi-layer tissues. With the developed hybrid bioink, the layered structures were fabricated successfully by using the printing and culturing system. Results indicated that the strand width could be 343±28um in single layer and there was regular and intact shape in layered structures; And PC12 cells were uniformly distributed in 3D grid structure and had a high viability. The printed layered structures and encapsulated PC12 cells indicated that the system had the ability to achieve the multi-layer tissues printing.
