The study combined methanol extraction with enzyme pretreatment to extract taxol from Taxus Chinensis var. mairei, which was optimized by orthogonal test. Under the optimum conditions which were extracting four times at 50°C for 10.5h, 80 mesh, solid-liquid ratio 1:12,the extracting yield of taxol was 0.0262%. While the organic solvent extraction was combinated with enzymatic pretreatment, the yield was increased to 0.0301% under the conditions which were pretreated by enzyme activity of 34U/g(as cellulase/pectinase ratio=2:1) for 1h. The results showed that by combining the methanol extraction method with the enzyme-pretreatment method, the rate of extracting taxol from Taxus were increased significantly, which provided a certain basis and reference for industrial production.
