To investigate the inhibitory effect of Sargassum polysaccharide (SP) on IBDV induced lymphocyte apoptosis in chicken bursa and its underlying mechanism. The effect of Sargassum polysaccharide on lymphocyte apoptosis in chicken bursa was observed through morphology, DNA gel electrophoresis, flow cytometry and ELISA assay. The mRNA expression in Fas gene was then detected by real time quantitative RT-PCR. Sargassum polysaccharide significantly inhibited lymphocyte apoptosis in chicken bursa, with positive relation to the concentration of SP. Apoptosis rate was greatly reduced according to detection by flow cytometry. Mitochondrial membrane potential and reduced cytochrome C releasing were enhanced greatly by SP in a dose-dependent manner. Standard curve for Fas gene plasmid was established with real time quantitative PCR, indicating negative relation between Fas mRNA expression level and concentration of SP. The result indicated that Sargassum polysaccharide inhibited lymphocyte apoptosis in chicken bursa and reduced oxidation injuries of immunocytes caused by ROS.
