The leptin gene G2548A polymorphism, serum levels of organochlorine pesticides, and the risk of cholesterol gallstones: a case-control study

The aim of this study was to evaluate the association of a single-nucleotide polymorphism (SNP) G/A in the promoter of the leptin (LEP) gene and blood levels of organochlorine pesticides (OCPs) with cholesterol gallstone risk among the Chinese population. Two hundred patients with cholesterol gallstones were enrolled in a 1:2 matched case-control study. Genotyping for the SNP was conducted on the LightCycler480 polymerase chain reaction (PCR) platform. Blood OCP levels were measured by gas chromatography, coupled with the electron capture technique. Dimensionality reduction and crossover analysis were employed to assess interactive effects. Conditional logistic regression revealed that the AA genotype of LEP G2548A SNP and high serum β-hexachlorocyclohexane (HCH), δ-HCH, and p,p-dichloro-diphenyl-trichloroethane (DDT) concentrations are risk factors of cholesterol gallstones (odds ratios 2.29, 1.25, 1.12, and 2.86, respectively). Analysis in the MDR software via the optimal six-factor interaction model showed that the high-risk group (which featured a gene–environment interaction) was at an 8.55-fold higher risk (P < 0.001) of cholesterol gallstones than the low-risk group was (no gene–environment interaction). Crossover analysis indicated a statistically significant interaction between the LEP 2548G>A mutation and OCPs (high serum p,p- dichlorodiphenylethylene and o,p-DDT levels) according to the multiplicative model. The homozygous AA mutation at the leptin gene G2548A site and the high OCP exposure level may be risk factors for cholesterol gallstones, and there is an interaction (according to the multiplication model) that can increase the risk of this disease.
