Identification and characterization of a novel sub-cellular GA transporter

The plant hormone gibberellin (GA) is fundamental for many aspects of plant growth and development. Movement of GA within plants has been documented and proven to be essential for proper plant growth, however little is known about the molecular mechanisms executing GA transport. Only a small number of transporters have been characterized as GA transporters in planta. We have identified a member of the NITRATE TRANSPORTER 1/PEPTIDE TRANSPORTER family (NPF) capable of transporting GA in a Xenopus oocytes transport system. npf mutants produce less interfascicular fibers and more sieve tube elements. Formation of interfascicular fibers is promoted by GA, this result suggests that the knockout of this npf member results in the reduction of GA availability in the plant, causing the GA/Auxin balance to tip in favor of auxin, which in turn promotes the upregulation of sieve tube element formation. The transporter is localized to the vacuole, providing the first evidence of subcellular GA transport activity in plants.
